The primary and secondary antibodies described above were used at experimentally optimized dilutions. WebPrepare PBMCs from normal or donor blood. Pre-warm the BD Cytofix Buffer in a 37°C water bath for 5-10 minutes before use. (Optional) Culture PBMCs in RPMI with 5% human serum at 37°C in a CO 2 incubator for 2 hours. Treat the cells with appropriate stimulators. Fix the cells immediately to maintain their phosphorylation state.
